It depends a lot on where a mis-spelling is found. For instance, I don't mind spelling errors while in chat, in forums or e-mail. When I am researching a medical topic for a patient or client, however, I tend to start tuning out of incorrectly spelled writing almost immediately. If one is going to publish a learned treatise, then it needs to be checked for spelling, proof-read, and probably reviewed before it gets printed and distributed. On the other hand, in this kind of medium, it is more the idea that needs to be conveyed over the need for exactly the correct spelling and grammar. Besides, sometimes we have very good thoughts expressed by people who do not use English as their main language, and may have a completely different alphabet. Just because someone is not a native of this country and does not use the language with expert facility does not mean they have invalid thoughts, or are in any way mentally below those of us who have proper 'Merican fetchins up."
